Methotrexate increases the activation and cytotoxicity of 5-fluorouracil: detected by 19F n.m.r. in vivo.

چکیده
In the rat Walker 256 carcinosarcoma the cytotoxicity of the anticancer drug 5-fluorouracil (FU) is thought to be mediated by the 5-fluoronucleotide (FNuct) FUTP. which interferes with RNA metabolism [ 11. Total FNuct can be detected in vivo, as a composite peak, by "F n.m.r. and we have found in the Walker 256 carcinosarcoma that decreased concentrations of FNuct (mainly FUTP) formed in vivo accompanied decreased cytotoxicity [ 21. Sequential treatment of Walker tumours with methotrexate (MTX) followed by FU (MTX-FU) is known to produce increased cytotoxicity compared with either drug alone or to the reverse schedule (FU-MTX) [3]. This effect may be due to MTX increasing intracellular pyrimidine ribosylpyrophosphate (PRPP) and thus increasing anabolism of FU to FNuct [4]. The aim of this study was to use "F n.m.r. to determine whether MTX pretreatment increases FNuct formation in vivo and to correlate this with cytotoxicity.

In vivo measurements of intratumoral metabolism, modulation, and pharmacokinetics of 5-fluorouracil, using 19F nuclear magnetic resonance spectroscopy.
In vivo 19F nuclear magnetic resonance spectroscopy was used to monitor and measure 5-fluorouracil and some of its cytotoxic anabolites directly in rats bearing the Walker 256 adenocarcinoma by using a 4.7-T horizontal bore magnet. A two-compartment subsystem model was used to estimate intratumoral transfer parameters. The apparent rate of formation of the nucleosides/tides in the tumor, K21, c...
متن کاملEffects of 5-fluorouracil on dihydrofolate reductase and dihydrofolate reductase mRNA from methotrexate-resistant KB cells.
Growth of methotrexate-resistant dihydrofolate reductase gene-amplified KB cells in the presence of 5-fluorouracil results in an increase in dihydrofolate reductase mRNA. This increase can be solely attributed to a species of RNA of approximately 3.5 kilobase pairs in size. Although dihydrofolate reductase enzyme activity increases per cell with increasing 5-fluorouracil, there is a decrease of...

In this study, seven-layer nanofiber structures consisting of polycaprolactone/ chitosan polymers loaded with methotrexate and 5-fluorouracil anti-cancer drugs, for controlled drug delivery, were produced and evaluated. For this purpose, the second, fourth and sixth layers were loaded with drug and placed between the drug-free layers. The surface morphology of drug-free and drug-loaded nanofibe...

Background: Mycosis fungoides (MF) is a cutaneous T cell lymphoma. There are only palliative treatments in advanced stages with no cure. Most chemotherapy regimens have temporary effects with various side effects. Objective: The objective of this study was to assess the efficacy of a new chemotherapy regimens in advanced stages of MF. This regimens takes advantage of synergistic effect of metho...

19F-n.m.r. spectroscopy was used to study the binding of 3',5'-difluoromethotrexate to dihydrofolate reductase (tetrahydrofolate dehydrogenase) from Lactobacillus casei. The benzoyl ring of the bound difluoromethotrexate was found to 'flip' about its symmetry axis, and the rate (7.3 x 10s3-1 at 298 K) and activation parameters for this process were determined by lineshape analysis ofthe 19F-n.m...
